Salmonella L. monocytogenes and E.coli O157 mPCR

Simultaneous identification of Salmonella spp., L. monocytogenes and E. coli O157 DNA by multiplex polymerase chain reaction (m-PCR).


Fast molecular detection methods could be more convenient, particularly, Salmonella spp., L. monocytogenes and E. coli O157 m-PCR detection kit, enabling amplification of three products in parallel in a single reaction tube, helps user to detect multiple food pathogens with a considerable reduction of time analysis. Moreover, the kit includes an internal amplification control that is necessary to guarantee that negative PCR detection results are due to the absence of pathogens' nucleic acids and are not false negative results (Hoorfar et al. 2004). The proposed kit, providing high sensitivity and specificity and including an amplification control system could represent an efficient tool for the rapid screening of food samples.

As a consequence of an increasing amount of food contaminations and growing consumer concerns, food safety has become an essential public health problem. Salmonella spp., L.monocytogenes and E.coli O157 have emerged as significant food-borne pathogens that pose a serious problem in many countries. Institutions all over the world are multiplying their efforts to ensure food safety (Wallace et al., 2000). The hygienic control of raw materials and finished products, normally accomplished by cultural microbiological methods, is now a main requirement of institutions and food industry. However, the prolonged time, needed by conventional methods to give a final diagnosis of contamination, often limits the effectiveness of control programmes. For this reason, rapid, sensitive, specific and reliable methods are needed to guarantee the microbiological quality of food. Fast molecular detection methods could be more convenient, particularly, Salmonella spp. L.monocytogenes and E.coli O157 m-PCR detection kit, enabling amplification of three products in parallel in a single reaction tube, helps user to detect multiple food pathogens with a considerable reduction of time analysis.

Salmonella spp., L.monocytogenes and E.coli O157 detection kit allows the detection of mentioned target species DNA using polymerase chain reaction (PCR). The kit contains reagents and enzymes for the specific amplification of a 155-bp region of Salmonella spp; a 173-bp region of L. monocytogenes; a 217-bp region of E.coli O157. This PCR assay is sensitive enough to detect up to ten cell per reaction. To check for possible PCR inhibition, an internal control, giving an amplicon of 112 bp, is also supplied.

 

Kit Contents:

- Multiplex PCR Mix: 1200 μl (2 x 600 ul) containing primers, dNTPs, 10X reaction buffer, MgCl2 and an internal control (plasmid DNA including specific primer complementary sequences)
- Positive control: 200 μl (bacterial DNA of target species)
- Hot-Rescue DNA Polymerase: 20 μl
PCR-grade Water: 1000 μl
