Integration of retroviral DNA is an obligatory step of retrovirus replication because proviral DNA is the template for productive infection. Integrase, a retroviral enzyme, catalyses integration. The process of integration can be divided into two sequential reactions. The first one, named 3′-processing, corresponds to a specific endonucleolytic reaction which prepares the viral DNA extremities to be competent for the subsequent covalent insertion, named strand transfer, into the host cell genome by a trans-esterification reaction. The monoclonal antibody is an aid in the reasearch against HIV-1 and in the diagnostic test development.

Description

Integration of viral DNA into a chromosome of the host cell is an essential step in the retroviral life cycle. This process is catalyzed by the viral enzyme integrase (IN) through 3 steps: first step, two nucleotides are removed from the 39 ends of the viral DNA (39-end processing); second step, the recessed 39 ends of the viral DNA are then joined to 59 staggered sites in the target DNA in a concerted cleavage and ligation reaction (DNA joining); last step, integration is completed by repair of the short gaps flanking the viral DNA intermediate and subsequent joining of the 59 ends of viral DNA to the target DNA. HIV integrase contains 3 domains, an N-terminal HH-CC zinc-finger domain which is partially responsible for multimerization, a central catalytic domain and a C-terminal domain. Both central catalytic domain and C-terminal domain have been shown to bind both viral and cellular DNA. No crystal structure data exists with Integrase bound to its DNA substrates. HIV-1 integrase functions as a dimer or tetramer. Additionally, several host cellular proteins interact with integrase and may facilitate the integration process.
