Description: Thymidylate synthase uses the 5,10-methylenetetrahydrofolate (methylene-THF) as a cofactor to catalyze the methylation of deoxyuridylate to deoxythymidylate. This function maintains the dTMP (thymidine-5-prime monophosphate) pool critical for DNA replication and repair. The enzyme has been of interest as a target for cancer chemotherapeutic agents. It is considered to be the primary site of action for 5-fluorouracil, 5-fluoro-2-prime-deoxyuridine, and some folate analogs. Among several polymorphisms within TYMS gene, a six-basepairs deletion in the 3UTR region named TYMS1494del it has been related to a DFS and OS decreased in patient treated with 5-FU in adjuvant treatment for colorectal cancer, through a mechanism involving the mRNA stability reduction.
